Notch extracellular domain is detected within Delta cells (green) indicative of trans-endocytosis. Supplemental Figure 1.  MTM1 and Rab5Q79L are in discrete domains on extensively dilated endosomes.  BHK cells were transfected with MTM1C375S-FLAG and Rab5Q79L.  Cells were permeabilized with saponin, fixed and stained as described in Materials and Methods. MTM1C375S-FLAG was detected with a mAb directed against the FLAG epitope and a FITC-conjugated donkey anti-mouse IgG.  EEA1 was stained with human anti-EEA1 and a Texas Red-conjugated donkey anti-human IgG.  Rab5 was localized using a rabbit pAb directed against Rab5 and Cy5-conjugated donkey anti-rabbit IgG.  Insets are enlarged 2.2 times.  Bar 5 µm.
