Notch receptors are clustered and trans-endocytosed by Delta ligand cells. Confocal micrograph of a Delta expressing cell (left) interacting with a Notch expressing cell (right). Following interaction with Delta (blue), cell surface Notch (yellow) is clustered at cell-cell interfaces. Notch extracellular domain is detected within Delta cells (green) indicative of trans-endocytosis. Endocytosis of ligand while bound to Notch may produce a force sufficient to pull Notch apart and activate signaling.

Dynamics of leptomycin B-sensitive neucleocytoplasmic flux of parathyroid hormone-related protein.
MHC Lam, B Henderson, MT Gillespie and DA Jans

Figure 2. FRAP analysis of the export of GFP-PTHrP from the nucleus and nucleolus in transiently transfected COS-1 cells.

Movie 2aii: This movie shows a COS-1 cell transiently expressing GFP-PTHrP and treated for 3h with 0.4 ng/ml leptomycin B. A region of the cytoplasm in this cell is photobleached for 10s and subsequent images were collected at 10 s intervals for 450s.

Figure 3. FRAP analysis of the import of GFP-PTHrP into the nucleus and nucleolus in transiently transfected COS-1 cells.

Movie 3aii: This movie shows a COS-1 cell transiently expressing GFP-PTHrP. A region comprising the nucleus and nucleolus of this cell is photobleached for 10s and subsequent images were collected at 10 s intervals for about 450s.
